The in situ and label-free analysis of nano-bio interactions in intact single cells at nanoscale remains challenging due to the complexity and dynamic changes of cellular structures and nanomedicines. Here we developed an analytical protocol based on synchrotron radiation X-ray microscopy techniques to overcome the challenges, and summarized an optimized workflow for sample preparation, beamline selection, data acquisition and analysis.
